بررسی اثر 5-آزاسیتیدین به‌عنوان ماده ضدمتیلاسیون DNA بر روی صفات زراعی، القای آندروژنز از طریق کشت بساک و بیان ژن DNA متیل‌ترانسفراز در بافت برگی ذرت (.Zea mays L)

بهینه‌‌سازی روش‌‌های درون شیشه‌ای برای تولید گیاهان دابل‌‌هاپلوئید ذرت نقش مهمی در برنامه‌‌های اصلاحی این گیاه دارد. در این مطالعه اثرات ماده 5-آزاسیتیدین بر روی صفات زراعی، کارایی القای آندروژنز و همچنین بیان ژن DNA متیل‌ترانسفراز (AF229183.1) در دو مرحله رشدی ذرت شامل مرحله 7 تا 8 برگی و مرحله گلدهی مورد بررسی قرار گرفت. آزمایش به‌صورت فاکتوریل در قالب طرح بلوک‌‌های کامل تصادفی در سه تکرار اجرا شد. دو ژنوتیپ ذرت (DH5 × DH7 و ETMH-82) به‌‌عنوان عامل اول و تیمار بذور ذرت با 5-آزاسیتیدین (0، 5، 10 و 100 میکرومولار) به‌‌عنوان عامل دوم در نظر گرفته شدند. بذور تیمار شده در مزرعه کشت گردیده و در طول مراحل رشد، صفات مختلف مورفولوژیکی و زراعی اندازه‌گیری شدند. در آزمایش کشت بساک، بساک‌های حاوی میکروسپور‌‌هایی در مراحل تک‌هسته‌ای میانی تا تک‌هسته‌ای انتهایی انتخاب و در محیط‌کشت پایه YPm حاوی 1 میلی‌گرم در لیتر D-2,4 و 2 میلی‌گرم در لیتر BAP کشت گردیدند. اثرات متقابل ژنوتیپ × سطوح 5-آزاسیتیدین برای همه صفات مورد مطالعه به‌جز تعداد دانه در ردیف بلال، عمق دانه، قطر بوته، تعداد برگ در بوته و تعداد بلال اختلاف معنی‌‌داری را نشان دادند. بیشترین میزان وزن هزار دانه در تیمارهای 10 و 100 میکرومولار و همچنین بیشترین عملکرد‌‌ دانه و عملکرد بیولوژیک در تیمار 100 میکرومولار 5-آزاسیتیدین برای هر دو ژنوتیپ مشاهده شد. بذور ژنوتیپ DH5 × DH7 تیمار شده با غلظت 5 میکرومولار 5-آزاسیتیدین بیشترین میانگین تعداد ساختارهای رویان مانند (1833/0) و گیاهچه باززایی شده (067/0) به‌ازای هر بساک را ایجاد کردند. بیان نسبی ژن DNA متیل‌ترانسفراز در گیاهان حاصل از بذور تیمار شده با غلظت‌‌های مختلف 5-آزاسیتیدین در هر دو ژنوتیپ و هر دو مرحله رشدی مورد مطالعه نسبت به‌‌ گیاهان شاهد (غلظت صفر میکرومولار 5-آزاسیتیدین) کاهش معنی‌‌داری نشان داد که این کاهش بیان ژن می‌‌تواند منجر‌به بهبود القای آندروژنز در کشت بساک ژنوتیپ DH5 × DH7 شده باشد. به‌هر حال، علی‌‌رغم کاهش بیان این ژن در دو مرحله رشدی ژنوتیپ ETMH-82، القای آندروژنز در این ژنوتیپ مشاهده نگردید. نتایج تحقیق حاضر می‌تواند به تعیین نقش عوامل اپی‌ژنتیکی در القاء آندروژنز و بهبود تولید گیاهان هاپلوئید در ذرت کمک کند.
