Abstract

Genetic diversity among Russian olive genotypes in three different regions of East-Azerbaijan province (includes Tabriz, Maragheh and Malekan) were assessed using morphological and molecular (AFLP) markers. Results of the quantitative and qualitative traits statistics showed a significant genetic variation among studied germplasm and categorized them in five distinguished groups. The most number of genotypes from Maragheh region were in group 1, while, the group 3 consisted of the majority of genotypes from Tabriz region. All genotypes were analyzed with 14 EcoRI-MseI primer combinations. A total of 439 informative and polymorph AFLP markers was generated and analyzed. Based on Jaccard Similarity Index, the minimum genetic similarity was observed between genotype 19 (from Malekan region) and 27 (from Maragheh). The principal coordinate analysis (PCoA) showed the suitable genomic distribution of AFLP markers among individuals. The highest (0.83 %) and lowest (0.33 %) polymorphic information content achieved by primers combination MTTT-EGA and MGT-ETA, respectively. Cluster analysis using molecular data and UPGMA algorithm, classified the studied genotypes in three distinguished groups. The genotypes at the same geographical region did not classify in the same group based on clustering by molecular data. However, this achieved to a large extent when they were classified groups using morphological traits. This result might be explained in terms of convergent evolution and some in part due to asexual propagation of Russian olive from the limited maternal genotypes and distribution across the different geographical regions.

Abstract

water stress and, in this regard, it is necessary to improve rice cultivars to tolerance to environmental stresses. In this research 154 recombinant inbred lines (F9) derived from a cross between Shah-Pasand and IR28 in three conditions (non-stress, osmotic stress -0.3 and -0.6 Mpa induced through polyethylene glycol-6000) were evaluated as a factorial experiment in randomized complete block design. In addition, for molecular polymorphism experiment, 110 SSR and EST-SSR markers were assessed on parents of population and among them, 41 markers identified which had proper polymorphism between two parents. The regression analysis between germination components and molecular markers revealed the most coefficient of determination were found in RM211 for allometric coefficient (17%) under non-stress, RMES10-1 for Plumule dry weight (18%) under -0.3 MPa; and RM273 for germination uniformity (22.7%) under -0.6 MPa. RM3496, RM452, and RMES6-1 in three conditions had the most number of significant relationships with six, three and eight traits, respectively, and they can be a suitable candidate for simultaneous improvement of several traits in breeding programs of marker-assisted selection. In addition, after the identification of significant markers associated with germination components, the closest genes to these markers were identified using bioinformatic analysis, and the analysis of their expression were performed by rice transcriptome database. According to the results, the maximum gene expression pattern under drought stress and under non-stress conditions were related to loci LOC_Os01g57220 and LOC_Os01g26039, respectively and this information could be applied in breeding programs.

Abstract

Tomato is the second most important vegetables, after potato, which has the highest area of cultivation worldwide. Acording to FAO statistics, Iran is the sixth producer of tomato cultivation in the world. However, more than 95 percent of vegetable seeds are imported into the country. The present study was conducted in order to evaluate commercial imported hybrids (8320, Eden, Matin and Xaman) and the F2 and F3 generations resulting from their self-breeding. In this regard, after the production of F2 and F3 generations, three generations (F1, F2 and F3) together with the control variety (Early Orbana y) were evaluated in the research farm of Seed and Plant Improvement Institute, Karaj, Iran, based on a randomized complete block design with four replications during 2019-2020. The analysis of variance showed that the difference among genotypes for single plant yield, days to flowering, average single fruit weight, number of fruits harvested and acidity (pH), was significant at the level of 1% probability and total soluble solids (TSS) was significant at the level of 5%. The results of the mean comparison showed that the highest mean of single plant yield belonged to 8320 F1 (10920/10g), the highest days to flowering was observed in Xaman F2 (47.01 days), the highest mean weight of single fruit was observed in Xaman F1 (115.50 g), the highest number of fruits harvested was in Xaman F1 (106.62), the highest amount of total soluble solids was in Matin F3 generation (4.96) and the lowest amount of acidity (pH) was in 8320 F1 (4.10). The results of estimating genetic parameters showed that most of the evaluated traits had high genetic and phenotypic variances, but a slight difference was observed between the coefficient of phototypic variation and the coefficient of genotypic variation, which indicates a small effect of environment on the control of these traits. Higher variance of dominance than additive variance in most traits caused the degree of dominance to be greater than one, Therefore, the possibility of control of traits by genes with dominance or over-dominance effects is high, and in order to improve these traits, the methods for hybrid development are suggested.
