بررسی تنوع ژنتیکی سرده انجیر (Ficus L.) در ایران با استفاده از نشانگرهای مولکولی توالی‌های تکراری ساده میانی

انجیر به عنوان یک محصول مهم، در چند دهه اخیر به دلیل بروز تنش­های زنده و غیر زنده دچار فرسایش ژنتیکی شده است. هدف از این تحقیق تعیین تنوع ژنتیکی گونه­های خودروی سرده Ficus در ایران با استفاده از نشانگر ISSR می­باشد. جهت انجام این تحقیق، تعداد 23 نمونه جمعیتی متعلق به  گونه­های این جنس از سراسر کشور جمع آوری و DNA ژنومی آنها از برگ استخراج گردید. از میان 18 آغازگر ISSR مورد آزمایش، شش جفت آغازگر که باندهای مناسب و قابل تحلیل داشت، انتخاب و فرآیند PCR پس از تعیین دمای مناسب برای آنها انجام شد. در مجموع تعداد 83 باند الکتروفورزی PCR تولید شد که از این تعداد 78 باند پلی­مورفیک بودند.  از میان آغازگر­های استفاده شده، آغازگرهای (AG)8 C، (TG)8 A و (GT)8 C مناسب‌ترین آغازگر برای کاربرد در مطالعات آتی تشخیص داده شد. بر اساس نتایج به دست آمده از دندرو گرام رسم شده حاصل از آنالیز داده­های ISSR، 23 جمعیت Ficus  در چهار گروه قرار گرفتند. نتایج حاصل از تحلیل واریانس مولکولی نشان داد که بالاترین میزان فاصله ی ژنتیکی بین جمعیت‌های درون گونه‌ها است. نتایج این تحقیق نشان داد که تاکسون های متعلق به سرده Ficus در ایران از لحاظ تبارزایی بسیار به هم نزدیک می­باشند و ارتباط تولید مثلی و جریان ژنی بین آنها بالا است.
