بررسی تنوع درون‌گونه‌ای و بین‌گونه‌ای گیاه فستوکا با استفاده از الگوی الکتروفورز پروتئین‌ها

فستوکا یکی از بزرگترین جنس‌ها از خانواده گراس‌ها است که بیش از 600 گونه با سطح پلوئیدی متفاوت دارد. این مطالعه با هدف بررسی تنوع ژنتیکی 22 ژنوتیپ از سه گونه فستوکا (Festuca arundinacea، F.ovina و F.rubra) با استفاده از الگوی الکتروفورز پروتئین‌های ذخیره‌ای بذر انجام شد. این گونه‌ها تنوع قابل‌توجهی در تعداد باند‌های پروتئینی از 13-5 نشان دادند. بیشترین تعداد باند در G17 (F.rubra) و کمترین تعداد باند پروتئینی در G5 (F.ovina) مشخص شد. باند شماره 14 کمیاب بود و فقط در G3 در گونه F.ovina مشاهده شد که می‌تواند به‌عنوان یک باند اختصاصی برای شناسایی این ژنوتیپ در نظر گرفته شود. با توجه به نتایج تجزیه AMOVA سطح بالایی از تنوع ژنتیکی درون‌گونه‌ها نسبت به بین‌گونه‌ها وجود داشت که می‌تواند ناشی از ماهیت دگرگشنی در این جنس باشد. با توجه به اختلاف مشاهده شده در شاخص‌های تنوع بین سه گونه مورد مطالعه، مشخص شد که گونه‌ها دارای ساختار ژنتیکی متفاوتی هستند. نتایج تجزیه خوشه‌ای بر اساس الگوی پروتئین ذخیره‌ای بذر در ژنوتیپ‌های ارزیابی‌شده با استفاده از ماتریس فاصله اقلیدسی و روش UPGMA، ژنوتیپ‌های مورد مطالعه را در چهار گروه قرار داد. کمترین ضریب تشابه بین G14 و G15 (F.arundinacea) با G6 (F.ovina) وجود داشت، لذا می‌توان نتیجه گرفت گونه‌ها از روند تکاملی متفاوت‌تری تکامل یافته‌اند و بنابراین توصیه می‌شود به‌عنوان والد در تولید ارقام ترکیبی استفاده شوند. تنوع مشاهده شده در الگوی پروتئینی بذر گونه‌های فستوکا می‌تواند به‌علت هتروزیگوتی ناشی از دگرگشنی، تفاوت گونه‌ها یا جمع‌آوری جمعیت‌ها از مناطق متفاوت باشد.
