بررسی خصوصیات کیفی و بیان ژن‌های دخیل در کیفیت نانوایی برخی از لاین‌های دابلدهاپلوئید گندم نان

کیفیت نان به کمیت و کیفیت پروتئین آرد گندم بستگی دارد و این مستلزم استفاده از ارقام مناسب گندم می‌باشد. هدف از این مطالعه بررسی کیفیت نانوایی لاین‌های دابلدهاپلوئید گندم و ارتباط بین خصوصیات کیفی با پروتئین‌های ذخیره‌ای گلوتنین بوده است. صفات مرتبط با کیفیت نانوایی 30 لاین دابلدهاپلوئید گندم به‌همراه والدین و همچنین دو رقم Ehsan و Morvarid به‌عنوان شاهد مورد ارزیابی قرار گرفتند. آزمون SDS-PAGE جهت مشخص نمودن میزان پروتئین کل و ارتباط بین پروتئین‌های ذخیره‌ای بذر با خصوصیات کیفی انجام شد و سپس آزمون بیان ژن‌های دخیل در کیفیت نانوایی صورت گرفت. نتایج نشان داد که بین ژنوتیپ‌های گندم اختلاف معنی‌داری در تمامی صفات مورد ارزیابی وجود دارد. بیشترین میزان حجم رسوب زلنی مربوط به لاین‌های DH-143 و DH-159 (به‌ترتیب با 34 و 31 میلی‌لیتر)، بیشترین گلوتن مرطوب مربوط به DH-159 (77.80 گرم) و DH-143 (74.85 گرم)، بیشترین گلوتن خشک مربوط به DH-159 (26.21 گرم) و DH-143 (25.11 گرم) و بیشترین میزان جذب آب مربوط به DH-159 (51.59 درصد) و DH-143 (49.74 درصد) بود. همچنین بیشترین محتوای پروتئین مربوط به DH-143 (18.03 درصد) و DH-159 (17.72 درصد) بود. بررسی خصوصیات کیفی نانوایی نشان داد که لاین‌های DH-143 و DH-159 مطلوب‌تر می‌باشند. نتایج آزمون SDS-PAGE مشخص نمود که بالاترین میزان پروتئین دانه مربوط به لاین‌های DH-143 و DH-159 (به‌ترتیب 28.23 و 26.63 میکروگرم در میلی‌لیتر) می‌باشد. بر اساس نتایـج حاصـل از آزمـون بیـان ژن (Real-Time PCR) مشخص شد که لاین‌های DH-159 و DH-143 میزان بیان بالاتری را برای ژن‌های HMW-X، HMW-Y و PDIL در مقایسه با رقم شاهد از خود نشان دادند. با توجه به نتایج حاصله از این مطالعه، لاین‌های DH-143 و DH-159 را می‌توان در برنامه اصلاحی برای بهبود کیفیت نانوایی گندم مورد استفاده قرار داد.
