ارزیابی تنوع ژنتیکی در بین نمونه‌های مختلف گیاه استبرق با نشانگرهای مولکولی ISSR

مطالعه حاضر جهت طبقه‌بندی و بررسی تنوع ژنتیکی بین نمونه­‌های استبرق مناطق مختلف استان کرمان با استفاده از نشانگرهای ISSR انجام شد. در مجموع DNA مربوط به 14 نمونه گیاهی با 9 آغازگر ISSR با استفاده از واکنش PCR تکثیر شد و الگوی باندی آن‌ها به‌دست آمد. آغازگرهای مورد استفاده چندشکلی قابل‌قبولی (35.93) را نشان دادند به‌نحوی‌که حداقل و حداکثر شاخص اطلاعات چندشکل آغازگرهای به‌کار رفته در این مطالعه به‌ترتیب 0.11 برای آغازگر ISSR9 و 0.41 برای آغازگرهای ISSR3 و ISSR8 بود. میزان شباهت ژنتیکی بر اساس شاخص نی از 0.405 تا 0.745 به‌دست آمد و کم‌ترین شباهت ژنتیکی بین نمونه‌های جیرفت 3 و دوساری 2 و بیشترین شباهت ژنتیکی بین نمونه‌های جیرفت 1 و جیرفت 2 بود. با استفاده از تجزیه خوشه‌ای به‌روش UPGMA، نمونه‌ها در چهار گروه قرار گرفتند که گروه‌های دوم و سوم نمونه‌های بیشتری را در خود جای دادند. از نظر نزدیکی ژنتیکی نیز دو نمونه جیرفت 1 و جیرفت 2 که در یک خوشه قرار داشتند، نزدیک‌تر بودند. همچنین نمونه‌های جمع‌آوری شده از عنبرآباد نسبت به سایر نمونه‌ها در فاصله ژنتیکی دورتری قرار داشت. تجزیه به مختصات اصلی نیز نشان داد مؤلفه‌های اول و دوم 67 درصد از تنوع به‌دست آمده را توجیه می‌کنند. به‌طور کلی نشانگرهای ISSR برای طبقه‌بندی نمونه‌های استبرق مفید بود و با توجه به اطلاعات به‌دست آمده مبنی بر وجود تنوع ژنتیکی در بین نمونه‌های استبرق استان کرمان، از این تنوع می‌توان در آینده در به‌نژادی و تولید استبرق زراعی بهره جست.
