بررسی تنوع ژنتیکی اکوتیپ‌های اسفرزه (Plantago ovata) با استفاده از صفات مورفو-فنولوژیک و نشانگرهای ISSR

اسفرزه (Plantago ovata) گیاهی است که برای کاهش التهاب معده و عفونت ادراری و همچنین کنترل قند خون و میزان کلسترول در بدن استفاده می‌شود. روابط فیلوژنی و تنوع ژنتیکی 22 اکوتیپ مختلف اسفرزه (Plantago ovata) با استفاده از 12 نشانگر ISSR و همچنین نه صفت مورفولوژیکی و فنولوژیکی مورد ارزیابی قرار گرفت. نتایج تجزیه واریانس داده‌ها حاکی از تنوع بالا بین اکوتیپ‌های مورد مطالعه بود. تجزیه خوشه‌ای به روش UPGMA توانست 22 اکوتیپ مختلف را بر اساس داده‌های زراعی در دو گروه قرار دهد. همچنین ارزیابی مولکولی اکوتیپ‌ها نشان داد که 12 آغازگر توانستند تعداد 91 نوار چندشکل به وجود آورند. از بین آغازگرهای مورد استفاده، آغازگر UBC813 با 11 نوار و بعد از آن، آغازگر UBC811 با تعداد 10 نوار بیشترین و آغازگر UBC824 با تعداد 4 نوار کمترین تعداد نوار چندشکل را ایجاد نمودند. محتوای اطلاعات چندشکل (PIC) نشانگرها بین 26/0 تا 45/0 و شاخص نشانگری (MI) از 90/0 تا 13/4 متغیر بود. تجزیه خوشه‌ای به روش UPGMA براساس داده‌‌های مولکولی، 22 اکوتیپ مورد مطالعه را در پنج گروه قرار داد که به ترتیب شامل 1، 1، 2، 3 و 15 اکوتیپ بودند. گروه‌بندی اکوتیپ‌ها با نشانگرهای مولکولی با گروه‌بندی اکوتیپ‌ها با صفات مورفولوژیک تفاوت داشت. با توجه به نتایج می‌توان از اکوتیپ‌هایی که فاصله زیادی با هم دارند در برنامه اصلاحی اسفرزه استفاده نمود.
