اثر براسینواستروئید بر عملکرد دانه، برخی از صفات فیزیولوژیکی و بیان ژن‌های مرتبط با مسیر سیگنالینگ این هورمون در گندم تحت تنش خشکی

به‌‌منظور بررسی اثر محلول‌‌پاشی غلظت‌‌های مختلف اپی‌‌براسینولید بر عملکرد دانه‌‌ای، کاتالاز، محتوای کلروفیل کل، پایداری غشاء سلولی گندم و بررسی بیان برخی از ژن‌‌های مسیر سیگنالینگ براسینواستروئیدها (BES1 و BRI1) تحت تنش خشکی انتهای فصل، آزمایشی به‌‌صورت کرت‌‌های دو بار خرد شده در قالب طرح بلوک‌‌های کاملاً تصادفی با سه تکرار در مزرعه تحقیقاتی مؤسسه تحقیقات اصلاح و تهیه نهال و بذر کرج در سال زارعی 98 به ‌‌اجرا درآمد. عامل اصلی در دو سطح (آبیاری کامل و قطع آبیاری از مرحله 50 درصد گلدهی تا پایان فصل زراعی)، عامل فرعی در چهار سطح (محلول‌پاشی اپی‌براسینولید در 0، 0.25، 0.625 و 1 میلی‌‌گرم در لیتر) و عامل فرعی فرعی شامل هفت ژنوتیپ‌‌ گندم (مهرگان، پارسی و ژنوتیپ‌‌های ناشناخته با کدهای 2853، 3506، 3737، 4056 و 4228) بود. نمونه‌‌برداری سی روز پس از اعمال تنش خشکی از برگ پرچم (مرحله زادوکس89) صورت گرفت. نتایج نشان داد که تنش خشکی موجب کاهش پایداری غشاء سلولی، محتوای کلروفیل کل، عملکرد و همچنین افزایش کاتالاز در همه ژنوتیپ‌‌ها می‌‌شود. ژنوتیپ 4228 بر اساس میزان عملکرد دانه‌‌، پایداری غشاء سلولی، محتوای کلروفیل و کاتالاز تحت تنش خشکی، متحمل‌‌ترین ژنوتیپ گندم در بین ارقام مورد مطالعه بود. همچنین، نتایج نشان داد که کاربرد اپی‌‌براسینولید از طریق افزایش صفات مذکور موجب کاهش اثرات مخرب تنش خشکی در گندم شده که نتیجه آن افزایش عملکرد دانه‌‌ای بیشتر تحت تنش خشکی است. با افزایش غلظت اپی‌براسینولید، میزان عملکرد دانه نیز افزایش یافت. بررسی الگوی بیان ژن 1TaBES و TaBRI1 با استفاده از روش Real-time PCR نشان داد که اگرچه کاربرد هورمون براسینواستروئید موجب افزایش تحمل به تنش خشکی در گندم می‌‌شود اما مسیر سیگنالینگ آن متفاوت از مسیر سیگنالینگ شناخته شده BRI1 است
