تأثیر تنش خشکی بر بیان ژن‌های درگیر در سنتز و هیدرولیز فروکتان در طی انتقال مجدد ذخایر فتوسنتزی ریشه گندم

در شرایط تنش خشکی که یکی از مهم‌ترین عوامل محدودکننده عملکرد گندم در مناطق خشک و نیمه‌خشک محسوب می‌شود، انتقال مجدد مواد فتوسنتزی به منظور پر کردن دانه‌های گندم اهمیت بیشتری می‌یابد. در مورد اهمیت ریشه در انتقال مجدد طی پر شدن دانه‌ها تحت تنش خشکی اطلاعات اندکی در دسترس است. لاین موتانت پیشرفته گندم نان (T-65-7-1) به همراه تیپ وحشی آن (رقم طبسی) در دو شرایط رطوبتی (مطلوب و 40-30 درصـد ظرفیت مزرعه) به صورت یک آزمایش فاکتوریل در قالب طرح کاملاً تصادفی در سه تکرار کشت شدند. نمونه‌برداری‌ برای آنالیز بیان ژن‌ها در 2 مرحله (7 و 21 روز پس از گرده‌افشانی) از ریشه انجام شد. در این ژنوتیپ‌ها انتقال مجدد و کارایی انتقال مجدد فروکتان و بیان نسبی ژن‌های درگیر در سنتز و هیدرولیز فروکتان در ریشه طی پر شدن دانه‌ها، تحت تنش خشکی انتهایی مطالعه شدند. نتایج نشان داد که فروکتان ذخیره شده در ریشه در انتقال مجدد آسیمیلات‌ها سهیم بود. بیشتر بودن انتقال مجدد فروکتان از ریشه به دانه، تحت تنش خشکی در لاین موتانت به دلیل بالاتر بودن بیان ژن‌های درگیر در سنتز فروکتان (1-SST و 6-SFT) در 7 روز پس از گرده‌افشانی و هیدرولیز فروکتان (6-FEH) در 21 روز پس از گرده‌افشانی، نسبت به تیپ وحشی بود. تنش خشکی باعث تغییرات معنی‌داری در بیان ژن‌های 1-FFT و 1-FEH در ریشه هر دو ژنوتیپ نشد که تأیید کننده این موضوع است که تنها فرم غالب پیوند فروکتان β (2,6) تحت تأثیر تنش خشکی قرار گرفته است. در برنامه‌های اصلاحی گندم، ژن‌های 1-SST، 6-SFT و 6-FEH می‌توانند به عنوان نشانگرهای مولکولی برای گزینش ژنوتیپ‌هایی با محتوای فروکتان بالاتر و انتقال مجدد بیشتر مورد استفاده قرار گیرند.
